Can I build a custom package that already has the speedtree trees in it, as well as other assets? Thanks.

I will say Yes. You can export a package with everything that is in your scene. If you import it back in to a new project, the speedtrees will most likely have to be regenerated.
It shouldn't be neccessary to regenerate the speedtree materials if the package was made with the same version of Unity to what the OP is using ie 5.2.3.
